Estudio de la regulación transcripcional del cáncer de mama en mujeres mexicanas con sobrepeso y obesidad por microarreglos. (Spanish)

Breast cancer is the leading cause of death in women. Currently, the signalling pathways and genes involved in carcinogenesis, as well as their relationship to adipose tissue and their consequent association with poor prognosis are not fully described. The objective was to identify key genes of the tumour process in breast cancer in overweight and obese Mexican women. Using microarrays, the transcriptional regulation of breast cancer tumour tissue was compared with that of the adjacent non-tumour tissue of the mammary gland. Subsequently, the bioinformatics analysis identified 174 genes with fold change ≥ or ≤ 2 and a p ≤ 0.05. 109 genes were overexpressed and 65 underexpressed in breast cancer tumor tissue in association with excess adipose tissue. This shows that in breast cancer tumour tissue associated with obesity there is deregulation of genes that modify the process of apoptosis. We propose that the HAAO gene involved in the process of NAD synthesis is key in the regulation of the NAD+/-SIRT1-p53 axis, and also highlight the impact of the overexpression of the RPS6KB2 gene in the FSH hormone signaling pathway. El cáncer de mama es la principal causa de muerte en mujeres. Actualmente las vías de señalización y los genes implicados en la carcinogénesis, así como su relación con el tejido adiposo y su consecuente asociación con un mal pronóstico no están completamente descritos. El objetivo fue identificar genes clave del proceso tumoral en cáncer de mama de mujeres mexicanas con sobrepeso y obesidad. Mediante microarreglos se comparó la regulación transcripcional de tejido tumoral de cáncer de mama con la del tejido adyacente no tumoral de la glándula mamaria, posteriormente el análisis bioinformático identificó 174 genes que presentaron fold change ≥ o ≤ 2 y una p ≤ 0.05; 109 genes fueron sobreexpresados y 65 subexpresados en tejido tumoral de cáncer de mama en asociación con exceso de tejido adiposo. Lo que evidencia que en el tejido tumoral de cáncer de mama asociado con condiciones de obesidad existe desregulación de genes que modifican el proceso de apoptosis. Nosotros proponemos que el gen HAAO implicado en el proceso de síntesis de NAD es clave en la regulación del eje NAD+/-SIRT1-p53, además se destaca el impacto de la sobreexpresión del gen RPS6KB2 en la vía de señalización de la hormona FSH.
