中草药提取物对溃疡性结肠炎的调节作用. (Chinese)

The study was to explore the regulatory effect and possible mechanism of Chinese herbal extracts on UC. Sixty SPF Kunming mice, half in male and female, were randomly divided into six groups, which were blank control group (BC), model group (Model), positive group (SASP), Chinese herbal extract high-dose group (ZY-H, 16 mg/g), Chinese herbal extract medium-dose group (ZY-M, 8 mg/g), and Chinese herbal extract low-dose group (ZY-L, 4 mg/g), with ten mice in each group. After successful modeling, the mice in each group were given the drug for 10 d. The results showed that compared with BC group, the content of TNF-α, IL-1β, IL-17, IFN-γ, MPO and MDA in th colon of group Model were extremely increased (P<0.01), while the content of IL-10 and PPAR-γ and the activity of SOD and GSH-Px were extremely decreased (P<0.01). Compared with group Model, DAI index and colonic histopathological score of mice in all drug treatment groups were extremely decreased (P<0.01). The content of cytokines TNF-α, IL-1β, IL-17, IFN-γ, MPO and MDA were extremely decreased in ZY group and SASP group (P<0.01), while the content of IL-10 and PPAR-γ, and the activity of SOD and GSH-Px were extremely increased (P<0.01). The study indicates that each dose of Chinese herbal extract has positive regulatory effect on improving UC symptoms and inflammatory cytokines and oxidative stress products in colon tissue of mice. 试验旨在探究中草药提取物对溃疡性结肠炎(UC) 的调节作用及可能作用机制。试验选取60 只 SPF 级昆明小鼠,雌、雄各半,随机分为6 组,即空白对照组(BC)、模型组(Model)、阳性组(SASP)、中草药 提取物高剂量组(ZY-H,16 mg/g)、中草药提取物中剂量组(ZY-M,8 mg/g)、中草药提取物低剂量组 (ZY-L,4 mg/g),每组10 只小鼠。造模成功后,各组小鼠给药10 d。结果显示,与BC 组相比,Model 组小鼠结 肠肿瘤坏死因子-α (TNF-α)、白细胞介素-1β (IL-1β)、白细胞介素-17 (IL-17)、干扰素-γ (IFN-γ)、髓过 氧化物酶(MPO)、丙二醛(MDA) 含量极显著升高(P<0.01),白细胞介素-10 (IL-10)、过氧化物酶体增殖物 激活受体γ (PPAR-γ) 含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px) 的活性极显著降低 (P<0.01)。与Model 组相比,各药物处理组小鼠疾病活动指数(DAI)、结肠组织病理学评分均极显著降低 (P<0.01);ZY 各剂量组及SASP 组结肠细胞因子TNF-α、IL-1β、IL-17、IFN-γ、MPO、MDA 含量极显著降低 (P<0.01),IL-10、PPAR-γ 的含量及SOD、GSH-Px 的活性极显著升高(P<0.01)。研究表明,各剂量中草药提 取物均对改善小鼠UC 模型症状以及结肠组织中炎性细胞因子和氧化应激产物具有正向调节作用。.
