A recent study conducted by the Chinese Academy of Sciences has provided new information about the mechanisms underlying linear ubiquitination and its implications in tumorigenesis and drug discovery. Linear ubiquitination is a type of ubiquitination that involves attaching a head-to-tail polyubiquitin chain to a substrate protein. Dysregulation of linear ubiquitination has been linked to various types of cancers, such as lymphoma, liver cancer, and breast cancer. The study highlights newly discovered linear ubiquitination-mediated signaling pathways, recent advances in the role of linear ubiquitin in different types of cancers, and the development of linear ubiquitin inhibitors.
