Researchers at the Laboratory of Experimental Cardiology have conducted a study on gene editing, specifically targeting the proprotein convertase subtilisin-kexin type 9 (Pcsk9) gene. The loss-of-function of this gene has been associated with significant reductions in plasma serum low-density lipoprotein cholesterol (LDL-C) levels. The researchers used extracellular vesicles (EVs) as a delivery vehicle for CRISPR/Cas9 ribonucleoprotein (RNP) targeting of the Pcsk9 gene in primary mouse hepatocytes. The study found that the administration of Cas9 EVs successfully inactivated the Pcsk9 gene, leading to a reduction in Pcsk9 mRNA and increased uptake of the low-density lipoprotein receptor and LDL-C. This research provides valuable insights for future experiments and potential applications of EV-mediated CRISPR/Cas9 RNP delivery targeting Pcsk9. [Extracted from the article]
