A preprint abstract from biorxiv.org discusses the role of EWS::FLI1 in Ewing sarcoma and its impact on drug sensitivity. The study suggests that EWS::FLI1 promotes the formation of R-loops, which leads to increased interaction between DHX9 and RNA polymerase II. Additionally, EWS::FLI1 prevents the resolution of TOP1 poisoning-associated R-loops. The accumulation of R-loops results in replicative stress, genome instability, and increased sensitivity to genotoxic agents. This research provides new insights into the mechanisms behind Ewing sarcoma and potential implications for its treatment. However, it is important to note that this preprint has not yet undergone peer review. [Extracted from the article]
